Description

Acetylcysteineisusedmainlyasamucolytic,whichprotectsagainstacetaminophenoverdose-inducedhepatotoxicitybymaintainingorrestoringhepaticconcentrationsofglutathione.

InVitro

N-acetylcysteinepreventsapoptoticDNAfragmentationandmaintainslong-termsurvivalintheabsenceofothertrophicsupportinserum-deprivedPC12cells.N-acetylcysteinealsopreventsdeathofPC12cellsandsympatheticneurons[2].N-acetylcysteinecausesdose-dependentreductionsinviABIlityinratandhumanaorticsmoothmusclecells[3].N-acetylcysteineactivatestheRas-extracellularsignal-regulatedkinase(ERK)pathwayinPC12cells.N-acetylcysteineprotectsneuronalcellsfromdeathevokedbywithdrawaloftrophicsupport.N-acetylcysteineincreasesnitricoxide(NO)releasefromprotein-boundstoresinvasculartissue.N-acetylcysteinepretreatmentofPC12cellsinterfereswithNGF-dependentsignalingandneuriteoutgrowth,anditissuggestedthatN-acetylcysteineinterfereswithredox-sensitivestepsintheNGFmechanism[4].

InVivo

N-acetylcysteine(150,300mg/kg)treatmentsignificantlyreduceslivertransaminasesinallgroupsoftreatment,mostlyingroupN-acetylcysteine300.LungglutathioneperoxidaseissignificantlyincreasesingroupN-acetylcysteine300(P=0.04),whiletheotheroxidationbioMarkersshownosignificantdifferences[1].N-acetylcysteineimprovescognitionof12-month-oldSAMP8miceinboththeT-mazefootshockavoidanceparADIgmandtheleverpressappetitivetaskwithoutinducingnon-specificeffectsonmotoractivity,motivationtoavoidshock,orbodyweight[5].

RatsarerandomLyallocatedintofivegroups:shamgroup(n=5),controlgroupwithIIR(n=8)andthreegroupswithIIRwhoaregivenN-acetylcysteineindifferentdosages:150 mg/kgintraperitoneally5 minbeforeischemia(n=8,groupN-acetylcysteine150),300 mg/kgi.p5 minbeforeischemia(n=7,groupN-acetylcysteine300),and150 mg/kgi.p5 minbeforeischemiaplus150 mg/kg5 minbeforereperfusion(n=7,groupN-acetylcysteine150 + 150).After4 hofreperfusion,theanimalsareeuthanizedbyexsanguinationfromtheaBDominalaorta.MCEhasnotindependentlyconfirmedtheaccuracyofthesemethods.Theyareforreferenceonly.
